Routes you've clinched in one sitting

The major part of the Mackenzie Highway, from the southern end to where the Yellowknife Highway turns off.
Very nearly all of I-84 western, having turned off at I-205 (Oregon).
I-5 from Bellingham to Dunnigan, more times than I want to think about.
I-80 from Chicago to San Francisco.
I-40 from Barstow to Oklahoma.
CA-1
US-101 from San Francisco to L.A.

I-39 - WI and IL segments separately before they were connected (many times)
I-43 - a bunch
I-76 (western) - a bunch
I-135 (Kansas) - a few
I-335 (Kansas) - once, eastbound
I-380 (Iowa) - twice
Mountain, Daniel Boone, & Pennyrile Parkways - once each
Kansas Turnpike - few times
OK - Turner Turnpike and Will Rogers Turnpike (on same day) - few times
Ohio Turnpike and Indiana E-W Toll Road (on same day) - westbound
IL - Jane Addams Tollway - a bunch

I do not know if this counts, but back in the 70's when I-78 did not have all of its segments completed, I did travel from present day Exit 41 in Watchung, NJ to Exit 3 near Phillipsburg, NJ as well as from Fogelsville, PA to its western terminus in one shot.  We used to go to Hersheypark every year and would use that particular route.

Back in those days, Exit 41 at Drift Road in Watchung was the eastern terminus as all points east (except the NJ Turnpike) were not constructed in addition to the Allentown section that had the gap from west of the old Schaefer Brewery to Exit 3 in Greenwich, NJ where you had to use US 22 between those points. until 1990.  In retrospect, that was I-78 as being orphaned from the rest of the system, you could not count the Newark Bay Extension as I-78 and the NJTA did not even sign it until the section from Newark to Union opened later in the 1970's.
Every day is a winding road, you just got to get used to it.

I-88 in Illinois is the only one I've clinched at one time. 

For I-66 I never did the segment inside the beltway at the same time as the rest.  For I-30 I missed the part west of I-35W.  I've done some very, very long chunks of 65, 70, 80, 90, and 94 at once but not clinched.

No US highways clinched in their entirety in one trip, though I have done the entire Indiana section of US 31 at one time.
Indiana: counties 100%, highways 100%
Illinois: counties 100%, highways 61%
Michigan: counties 100%, highways 56%
Wisconsin: counties 86%, highways 23%

without even stopping for gas?  probably about 340 miles of I-10 in west Texas.  I may have even had cruise control on 84mph the whole way.

for entire routes, I think the longest on one tank of gas is probably CA-62.  that's only about 150 miles, though. 

I did I-40 once but took two rest breaks, countless gas stops, etc.

Criteria were not very clear.  If you can get off the highway to sleep for one night at someplace adjoining, how about for two nights as long as you stay within sight of the highway?  Obviously I didn't do all of those in one day.

I did I-27 with no overnight stop. I-57 all in one go, but with an overnight stop in Effingham (more like an effing pain in the ass).

My interpretation of the OP's criteria is that you can stop to use the bathroom, eat, get gas, or sleep, so the trip could last for a few days and still count, but you mentioned two nights in the same place.  If there was a day of tourism, visiting relatives, etc., then I would count it as two separate trips and not one.

No, I would count this as an all-at-once clinch. Say you hit I-68 at Morgantown and drive to Cumberland, and stay two nights. Then you get back on at the same exit and continue east to Hancock. That's one trip.
Government would be tolerable if not for politicians and bureaucrats.

I'm struggling to come up with any route I've clinched in one sitting that doesn't count as "short".  I'll throw I-27 out there; maybe that counts as "long".

I almost got eastern I-84 a few years ago, all but a bit on the west side of Hartford CT.

2-digit Interstate segments by state:
39 IL
70 IL, IN
71 KY
75 KY
77 VA
80 NE
84 PA, NY, MA
90 WI, IL
95 GA, NH

Other:
I-271 Cleveland
I-470 Wheeling
I-670 Columbus many times
OH 315 a few times

I should note that I don't usually go out of my way to clinch routes or counties.  Also, a couple of the items I listed were achieved as a young passenger on a family road trip, though there are probably more of those I can't remember at the moment.

There are several, but the ones that come to mind are: in MI US-8, M-14, I-275, M-36, M-22, M-108 <defunct>, M-120; In OH I-280, I-75.  I know there's more, but I'm not putting the effort into thinking right now.

I was referring to entire routes, not just routes through one state.

And not referring to short state routes, either. I clinch KY 498 (2.5 miles) several times a week. I was referring to through Interstate routes (one- and two-digit) and US highways.

I have never clinched a US Federal Route.  :-D

In February, I clinched a Mexican Federal Route (the Mex 2D toll road in northern Baja California), in one trip.

Some tiny Interstates can be clinched in about one or two minutes, Interstate 790 in Utica, NY, Interstate 587 in Kingston, NY, Interstates 175 and 375 in St. Petersburg, FL are four examples! :-D

Non-trivial routes. I'd set the bar at an hour. That's why I only included the longest beltways in my list.
