There are two segments of CR 280 in Walton County. The western section extends east from Bob Sikes Road and Eglin Air Force Base 6.09 miles to U.S. 331 at DeFuniak Springs. The eastern portion angles southward from U.S. 90 and 2nd Street in DeFuniak Springs to Eucheeanna and CR 183.

Within Washington County, CR 280 runs 15.46 miles east from CR 284 at Hinsons Crossroads to Whitehead Crossroads, Five Points, Johnson Crossroads and SR 277. The succeeding branch eastward follows Brickyard Road for 5.66 miles across Chipley between SR 277 and Sewell Farms Road. CR 280 also stems east from CR 273 (Orange Hill Road) 4.21 miles along Corbin Road to the Jackson County line, where it extends another 2.70 miles to U.S. 231 at Steele City.

County Road 280 in Jackson County travels east from CR 167 (Fairview Road) along Thompson and Magnolia Roads 6.91 miles to SR 71 at Oakdale. CR 280 continues southeast from SR 71 along Rocky Creek Road for 7.18 miles to CR 275 (Church Street) south of Cypress. A separate branch of CR 280 joins CR 69A at Shady Grove with CR 286, 4.01 miles to the east near Sinai.

Bob Sikes Road meanders 10.9 miles east through Eglin Air Force Base from SR 285 to Harrell Road.
