
Delaware 6 continues Maryland 291 eastward into Delaware by way of Millington Road. Millington Road carries Delaware 6 eastward to junction Delaware 42 (Blackiston Road) and the merge with Delaware 15 (Alley Corner Road) outside of Clayton (pop. 1,273). 1.3 miles after their merge, Delaware 15 parts ways with Delaware 6 (Millington Road) via Duck Creek Road in Clayton. Millington Road becomes Main Street through the heart of town where the state route crosses the Norfolk-Southern Railroad line.
East of Clayton, Delaware 6 (Smyrna Clayton Boulevard) joins Delaware 300 (Wheatleys Pond Road) for a 1.1-mile overlap into the town of Smyrna (pop. 5,679). The multiplex ends where Delaware 6 turns southward along U.S. 13 (Dupont Boulevard). Delaware 300 ends there and Delaware 6 shares three blocks of pavement with Dupont Boulevard before resuming an eastward course via Commerce Street.
Delaware 6 (Commerce Street) crosses over the SR 1 Turnpike and becomes Woodland Beach Road east of Smyrna. The state highway travels 5.2 miles between U.S. 13 and junction Delaware 9 (Hay Point Landing Road) at the Woodland Beach Wildlife Area. From there Delaware 6 continues three more miles along a causeway to the community of Woodland Beach along the Delaware Bay. During astronomical or storm related high tides the causeway portion of Woodland Beach Road is prone to flooding.

Delaware 6 ends at the riverside hamlet of Woodland Beach. There is no end sign posted and the highway itself ends at a boat ramp to the Delaware River and its associated parking lot.
