Business Loop I-40 at Flagstaff constitutes a 9.85 mile long route overlaying Historic U.S. 66 with the exception of the easternmost 0.77 miles.

The business route separates from Interstate 40 at a folded diamond interchange (Exit 191) near Belle Spring. Passing south of Observatory Mesa advances through the residential west side of Flagstaff to become a commercial boulevard. Business Loop I-40 converges with the north end of SR 89A by the campus of Northern Arizona University.

Turning northeast along Milton Road, Historic U.S. 66/Business Loop I-40 proceed into Downtown Flagstaff. U.S. 180 ties into the route from Humphreys Street south by Flagstaff City Hall and Colton Park. U.S. 180 accompanies Historic U.S. 66 east as Business Loop I-40 continues past Switzer Canyon and McMillan Mesa to East Flagstaff. This stretch also was originally part of U.S. 89.

A pair of trumpet interchanges and an access road previously linked U.S. 180, the east end of Business Loop I-40 and the south end of U.S. 89 with Interstate 40 at Exit 201. This changed with the grade separation at Business Loop I-40 and U.S. 89/180 eliminated and the junction at I-40 converted to a diamond interchange with access south to Country Club Drive. Historic U.S. 66 splits with Business Loop I-40/U.S. 180 ahead of the southward turn onto the viaduct spanning the BNSF Railroad and Industrial Drive to I-40.

U.S. 89 was dropped from a half mile overlap with Business Loop I-40/U.S. 180 between Santa Fe Avenue and the exchange with Interstate 40 with approval by the American Association of State Highway and Transportation Officials (AASHTO) on November 16, 2012. Associated action by AASHTO approved a request from the Arizona Department of Transportation (ADOT) to relocate U.S. 180 from Business Loop I-40 and SR 89A (Milton Road) southwest from Downtown Flagstaff to I-17/40, and along I-40 east to U.S. 89. The new alignment took U.S. 180 east along all of Business Loop I-40 east from Humphreys Street in Flagstaff to I-40.
