U.S. 62 extends 101.58 miles north into Western New York from the town of Kiatone in Chautauqua County to Niagara Falls.

U.S. 62 heads north from Warren, Pennsylvania to NY 60 southeast of Jamestown. U.S. 62 turns east to Frewsburg in the town of Carroll and then north to Interstate 86 in the town of Poland. Prior to 1972, U.S. 62 followed Akeley Road north from Akeley, Pennsylvania onto CR 53 (Warren Road) between Fentonville and Water Street in Frewsburg.

A stair stepped course takes U.S. 62 northeast to Ellington, Conewango and Leon. North from Dayton, U.S. 62 enters the village of Gowanda, where it combines with NY 39 across Cattaraugus Creek into Erie County. The two routes separate at Collins, with NY 39 turning east to Springville and U.S. 62 advancing north to the village of North Collins and NY 249.

Beyond the town of Eden, U.S. 62 continues into the town and village of Hamburg. South Park Avenue leads the route north past Buffalo Raceway and across I-90 (New York State Thruway) to U.S. 20 and the village of Blasdell.

U.S. 62 passes through the city of Lackawanna north to the Buffalo city line at South Park. The route angles northwest along South Park Avenue to Bailey Avenue, which takes U.S. 62 across both Cazenovia Creek and the Buffalo River to NY 16, I-190 and NY 354. U.S. 62 passes through the Lovejoy District, Schiller Park and the Kensington neighborhoods in east Buffalo.

Leaving Buffalo beyond the University Heights District, U.S. 62 (Bailey Avenue) leads north to Eggertsville within the town of Amherst. U.S. 62 shifts west along an overlap with NY 324 (Sheridan Drive) onto a commercialized stretch of Niagara Falls Boulevard.

Niagara Falls Boulevard arcs northwest around the city of North Tonawanda to the town of Wheatfield and Niagara Falls International Airport (IAG). Turning southwestward into the city of Niagara Falls, U.S. 62 comprises a five lane commercial boulevard to Interstate 190.

West of I-190, U.S. 62 (Niagara Falls Boulevard) partitions into a couplet following Walnut Avenue west and Ferry Avenue / Packard Road east. U.S. 62 Business branches one block north of U.S. 62 (Walnut Avenue) along Pine Avenue. The two routes run west to NY 104 (Main Street) at the Niagara Falls Business District.
