Business Loop I-25 follows a combination of U.S. 20-26-87 Business through the city of Casper. The route partitions at the south end with the northbound direction departing from I-25 at Exit 186 to Beverly Street south to Yellowstone Highway west. The southbound direction travels wholly along Yellowstone Highway east to its direct on-ramp with I-25.

Yellowstone Highway angles Business Loop I-25 & U.S. 20-26-87 Business southwest from C Street to 1st Street. 1st Street continues the four routes west to Center Street in Downtown Casper. There Business Loop I-25 & U.S. 87 Business turn north onto Center Street for the return to I-25. Locally maintained by the city of Casper, Center Street is former Wyoming 255.

Similar to the other end, the north end of Business Loop I-25 separates traffic northbound onto F Street, a freeway frontage road, west across the North Platte River to the on-ramp at Poplar Street (WYO 220). Business Loop I-25 south joins Center Street directly from the off-ramp at Exit 188A.
