State Road 46 travels 2.51 miles east from U.S. 441 at Mt. Dora to SR 453 (Mt. Dora Connector) and 46.16 miles from CR 46A near Mount Plymouth to U.S. 1 at Mims. With the relinquishment of 3.99 miles of SR 46 between SR 453 west of Sorrento and CR 46A, SR 46 is a split route. Further deletion of SR 46 will take place upon completion of SR 429 along Wekiva Parkway between southeastern Lake County and I-4 at Sanford. SR 429 opened to traffic west from I-4 westbound at Sanford in October 2022.

Lake County Road 46 spurs 0.60 miles west from U.S. 441 (Orange Blossom Trail) to Old County Road 441 (CR 500A) east of Downtown Mt. Dora. CR 46 was also designated on what was SR 46 between SR 453 and CR 46A. The SR 46 overlap with U.S. 17/92 (French Avenue) through Sanford is unsigned; instead trailblazers are posted at each end for the continuation of SR 46.

Construction associated with Sections 3A and 3B for the Wekiva Parkway expanded SR 46 east from U.S. 441 at Mt. Dora to a six lane arterial. Started on October 30, 2017, work on Section 3B removed the diamond interchange at the west end of SR 46 and added a two lane flyover from U.S. 441 south to the state road east. Section 3A widened 1.4 miles of SR 46 east from Section 3B to the exchange with SR 453 (Mt. Dora Connector). Both projects ran through Fall 2020.

The opening of the Mt. Dora Connector (SR 453) northwest from Orange County to SR 46 west of Sorrento on March 31, 2018 preceded the decommissioning of SR 46 through Sorrento and Mt. Plymouth. A portion of the route was repurposed as a frontage road.

Section 6 construction of Wekiva Parkway east from near Mt. Plymouth to Longwood Markham Road in Seminole County directly overlaid three sections of SR 46. The design-build project commenced on June 29, 2017 and wrapped up ~~Spring 2021~~ ~~early~~ Summer 2022. The parkway lanes opened on May 16, 2022. Further east, Section 7A upgraded existing SR 46 into the Wekiva Parkway mainline and added a frontage road system east to Orange Boulevard (CR 431). Section 7A work commenced on April 1, 2018 and ended in Fall 2022.

Wekiva Parkway branches southeast from SR 46 at Orange Boulevard along Section 8 (started on October 26, 2018 and underway through ~~Spring~~ Fall 2023) to connect with Interstate 4 at the Seminole Expressway (SR 417). Projected to run for 24 months, Section 7B construction began in Summer 2019. Widening the remaining 1.87 miles of SR 46 to six lanes east from SR 429 to I-4, work ran through ~~Spring~~ July 2022.

Martin Luther King, Jr. Boulevard was constructed from 2006 to 2008 as a new four lane arterial replacing Airport Boulevard (CR 425) as the main through route north from H.E. Thomas, Jr. Parkway (CR 46A) to SR 46 in Sanford. 02/29/08

Martin Luther King Jr. Boulevard intersects SR 46 across from Rand Yard Road. SR 46 comprises a four lane, divided arterial west to Seminole Towne Center mall and I-4 and east to U.S. 17/92 (French Avenue) outside Downtown Sanford. 02/29/08

French Avenue (old U.S. 17/92) south at SR 46 (West 1st Street) in Sanford. Sign changes made in 2018 realigned U.S. 17/92 westward from French Avenue alongside SR 46. 09/02/19
