Within the Palmetto State, U.S. 321 branches north from U.S. 17 at Hardeville in the LowCountry. U.S. 321 vacates South Carolina north of Clover to Gastonia, North Carolina.

U.S. 321 South Carolina Guides

U.S. 321 runs alongside a CSX Railroad corridor north from S.C. 119 near Scotia to Estill, Luray and Fairfax. The two continue northward to Sycamore and Ulmer, where U.S. 301 ties in from Allendale to the southwest. U.S. 301/321 separate just into Bamburg County, with U.S. 301 shifting eastward to Bamberg and U.S. 321 again traveling alongside the CSX Railroad to Olar, Denmark and Norway.

Continuing into Lexington County, U.S. 321 travels through Swansea and Gaston. U.S. 21/176 combine with U.S. 321 along Charleston Highway north from near Dixiana into the city of Cayce. The three routes line Knox Abott Drive east to Blossom Street across the Congaree River into Columbia.

U.S. 21/176/321 circumvent Downtown Columbia north along Huger Street to I-126/U.S. 76 and east on Elmwood Avenue to Main Street. U.S. 176 eventually branches northwest to St. Andrews, followed by U.S. 21 northeast to Blythewood. U.S. 321 continues northward to Winnsboro in Fairfield County, where the first of three business routes is designated.

U.S. 321 Business loops 4.35 miles east from U.S. 321 into Winnsboro. U.S. 321 Business in Chester follows Columbia Street and Center Street/Road along a 3.67 mile route through Downtown. North into York County, U.S. 321 passes through McConnells to the city of York, where a 2.50 mile long business route loops to the east.
