Interstate 335 follows the Kansas Turnpike northeast from Emporia, where I-35 branches east, to I-470 on the south side of Topeka. Traversing open agricultural or ranch land, there is a lone interchange along Interstate 335. Continuing the exit numbering system from the Kansas Turnpike north from I-35, Exit 147 joins the Kansas Turnpike with U.S. 56 in Lyon County, east of the small city of Admire.

The 50.17 mile long section of the toll road was added to the Interstate Highway System with concurrence by the American Association of State Highway and Transportation Officials (AASHTO) on December 5, 1987 to

facilitate the enactment of the new 65 mph speed limit on that portion of the Kansas Turnpike. All other segments of the Kansas Turnpike are currently classified as interstate routes.
