U.S. 6 runs 373 miles across the width of Nebraska from the Colorado state line in rural Chase County to the Glenville Dodge Memorial Bridge spanning the Missouri River between Omaha and Council Bluffs, Iowa.

Entering the Cornhusker State from Holyoke, Colorado, U.S. 6 travels to the Chase County seat of Imperial. Shifting southward from a 9.53 mile long overlap with N-61, U.S. 6 continues to Wauneta, Hamlet and U.S. 34 in Hitchcock County. U.S. 6/34 combine east from ahead of Cullbertson for 140.30 miles to Hastings.

Within McCook, U.S. 83 joins U.S. 6/34 along B Street for 1.44 miles. U.S. 6/34 meet U.S. 283 at Arapahoe and the west end of U.S. 136 north of Edison. Within Harlan County, N-4 branches east from the northern turn of U.S. 6/34 to Atlanta and U.S. 183 at Holdrege.

Paralleling a BNSF Railroad line, U.S. 6/34 bee line east from Holdrege to Funk, Axtell and Minden. Resuming an eastern heading into Adams County, U.S. 6/34 converge with U.S. 281 in the city of Hastings. U.S. 34 separates from U.S. 6 alongside U.S. 281 (Burlington Avenue) north into Downtown while U.S. 6 encircles the residential street grid along J Street and Elm Avenue.

Continuing from South Street in Hastings, U.S. 6 enters Clay County ahead of a 2.97 mile long overlap with N-14. U.S. 6 rejoins the BNSF Railroad line from Sutton east to Grafton and U.S. 81 at Fairmont in Fillmore County.

N-15 combines with U.S. 6 east ahead of Dorchester along a 11.82 mile long concurrency leading north into Seward County. N-15 branches north to Seward while U.S. 6 dog legs east to Milford and then north to L-80H. U.S. 6 parallels Interstate 80 from L-80H east into Lincoln.

U.S. 6 circumvents Downtown Lincoln along Sun Valley Boulevard and follows Cornhusker Highway, an arterial route through industrial areas through north Lincoln. U.S. 6 meets I-80 at Waverly and continues north of the freeway to Greenwood, Ashland and N-31 at Gretna.

U.S. 6/N-31 overlap 11.61 miles north along the suburban outskirts of the Omaha metropolitan area to the freeway along L-28B (W Dodge Road). N-31 continues north to Elkhorn while U.S. 6 overtakes L-28B along W Dodge Road east through west Omaha and Boys Town.

The West Dodge Expressway commences along U.S. 6 east from the exchange joining West Dodge Road with S 120th Street. U.S. 6 elevates onto a pair of three lane viaducts east to Interstate 680 while frontage roads connect with area streets and businesses below. U.S. 6 transitions into an urban boulevard east from Westroads Mall and I-680 to Midtown Omaha.

Heading east to Downtown Omaha, U.S. 6 partitions ahead of I-480/U.S. 75 along the couplet of Douglas Street east and Dodge Street west. U.S. 6 combines with I-480 east of S 10th Street across the Missouri River to I-29 in Council Bluffs, Iowa.
